研究概要 |

2003年4月にヒトゲノム塩基配列の完全解読が達成され、その後モデル動物として重要なマウスやラットのゲノムの塩基配列も決定され、時代はまさにポストシーケンスの時代、機能ゲノム学の時代に突入しています。遺伝子の塩基配列が決定されても、ゲノム上のどの部分が本当の遺伝子であるかは、まだ完全には同定されていません。ましてすべての遺伝子の機能の解明は、まだまだ今後の課題であります。当部門では、DNAマイクロアレイなどを用いたゲノムワイドな解析と、遺伝学的なアプローチから疾患遺伝子の同定や病態解明を行っています。ゲノム科学部門の研究の一つの大きな柱は、脂肪細胞分化あるいは骨芽細胞分化時の転写制御ネットワークの解明ならびに、それらの分化におけるクロストークの解明です(図1)。これらの転写制御ネットワークを解明することにより、メタボリック症候群や骨粗鬆症などの生活習慣病の病態解明ならびに、これらの病態改善のための創薬標的の描出あるいは診断マーカーの開発を目指しています。また、脂肪細胞分化における鍵転写因子であり、糖代謝、脂質代謝において創薬標的として重要な核内転写因子Peroxisome Proliferator-Activated Receptor(PPAR)を中心に新たな創薬標的の発見を目指し、またこれらの転写因子が制御する遺伝子を中心とした病態解明に力を入れています。
ヒトの病態を解明するために、モデル生物のゲノム・トランスクリプトーム・プロテオーム・フェノームなどの大量の情報を効率よく解析し、比較ゲノム学を用いた解析からヒトの病態に迫る研究も積極的に展開することにより、基礎研究と臨床研究の橋渡しを目指しています。
